The Diversity Outbred (DO) population is a heterogeneous stock of mice derived from eight genetically diverse founder strains. DO mice display a broad range of phenotypes; they also present new opportunities and challenges for systems genetics analysis. The talk will focus on mediation as a tool for exploring causal networks of gene expression and protein regulation. We will illustrate general principles of causal network analysis and highlight features that are unique to the DO as a model system. The availability of the Collaborative Cross – inbred strains derived from the same eight founders – provides a resource for independent validation of findings from the DO and presents new opportunities to advance our understanding of basic biology using the mouse as a model system.
